Phase 1 : In Silico Validation
- Objectif
- Determine whether the proposed H₂O₂ spatiotemporal gradients are physically achievable within OMIS, and whether the predicted non-monotonic signaling responses are plausible given known kinetic parameters.
- Coût estimé
- €500-2000
- Durée estimée
- 4-6 weeks
- Critères de réussite
- Surface H₂O₂ concentration after 10 min at 300 µL/min, 200 µM inlet · ≥100 µM (≤50% decrease) · (COMSOL simulation output at construct surface center point)
- Predicted PGC-1α fold-change for intermittent vs. sustained at 200 µM, 150 µL/min · Intermittent: 1.5–2.5; Sustained: 0.5–0.8; with ≤20% coefficient of variation across LHS · (ODE model output, mean ± SD from 100 LHS runs)
- Predicted shear stress magnitude at 150 µL/min · ≤0.5 Pa (below threshold for mechanotransduction in C2C12, per DOI:10.1007/s10439-012-0555-2) · (COMSOL wall shear stress at construct surface)
- On continue si
- All three success criteria met: surface concentration ≥100 µM, intermittent PGC-1α > sustained with non-overlapping confidence intervals, shear stress ≤0.5 Pa.
- On arrête si
- Surface concentration <100 µM at 300 µL/min, OR intermittent and sustained PGC-1α predictions overlap within 95% CI, OR shear stress >0.5 Pa.
- On réoriente si
- If surface concentration <100 µM but >50 µM, test higher flow rates (up to 500 µL/min) in simulation to see if laminar flow holds (Re < 100). If shear stress >0.5 Pa, redesign OMIS channel geometry (wider, shallower) to reduce shear.
- Risques
- H₂O₂ degradation rate in 3D construct is unknown and may be higher than literature values for 2D monolayersProbabilité : mediumParade : Use a wide range (k = 0.01–0.1 s⁻¹) in LHS; if no parameter set meets criteria, flag as high-risk and proceed to Phase 2 with direct measurement
- ODE model may not capture feedback dynamics (e.g., Nrf2-induced catalase upregulation within 10 min)Probabilité : mediumParade : Include time-delayed Nrf2 activation (τ = 5–15 min) based on DOI:10.1016/j.redox.2020.101678; if model fails, simplify to linear response assumption
- COMSOL license not availableProbabilité : lowParade : Use open-source alternative: OpenFOAM (v10) with reactingFoam solver, or FEniCS (v2019.2) with advection-diffusion-reaction module
